We have about 500 course on Totara, I would like to know who are the teachers and who are the editing teacher. Is there a readymade report.

There isn't a report source that provides information on who has a particular role in a course.
There is under Bulk user actions the ability to filter by course role when you select the show more. You can then disply or download these users.

Building on from this, is there a way to see where these permissions are? I can see who has the permissions by using your above method, but I have no idea which course they have these permissions in. I want to delete the role as I can see it's only used 4 times throughout the whole site, but I want to know which course they have the role in so I can assign a more appropriate role.

Solved my own problem! Went to the user's account, clicked Preferences then This User's role assignments. This showed me exactly what role the user had in which course.
