Hi there, is it possible for a grader to receive a notification not only when a learner submits an assignment (by file submission or online text), but also when he/she comments on the assignment? And vice versa, for the learner to receive such a notification when someone replies via the submission comments?

A notification around assignment comments is not an available option
There are events that can be monitored and subscribed to when they occur but assignment comments is not one of the events. if you go to Server > Events list and search for assignments the various rules show.

The event monitoring can be disbled by going to Quick-access menu > Server > Event monitoring rules.

Sorry, I meant: Is it possible to deactive the assignment comments function? If we can't receive a notification when someone comments we'd rather not make this available for the learners to use.

There is an Assignment setting under feedback types to disable marker comments

Many thanks. I've tried this and I'm afraid this disables the feedback comments for the markers, i.e. the marker is no longer to leave feedback comments on the learners submission and thus inline comments are also disabled.
Both the marker and the learner can, however, still post submission comments though.
Is there maybe a global setting that disables this for all assignments?

Unfortunately the only global settings for feedback is allow full anonymous and maximum days since completion.
There is some functionality iif you are using Perform with performance activities to get feedback on created question.
