We have been looking at the Totara/Moodle comparison chart and see that Totara supports custom landing pages for each system role.
Can anyone expand on this? What does this mean exactly and how is it setup. Is it simply different login pages, or is it a different front end/theme to the site all together?

I think that is referring to the role-specific dashboards - e.g. My Team for managers, My Learning for students. Originally we were going to have one for admins and one for trainers too.

I'm afraid you can't at the moment. The way dashboards were written does make it possible that we could extend it in that direction, but right now the learner and manager dashboards are the only ones available.
You can modify the contents of the default dashboards though - see Site Admin > Plugins > Dashboards > Manage Dashboards.
