Hi Jen,
Normally if you assign another role it won't remove the original role, and if you assign a role at a course level you should retain access at a site level.
The only circumstances that I can think of where this could happen is if you assigned the learner role in the system context AND you had modified the learner role and set a whole lot of capabilities to 'prohibit'.
The only way to fix this now is to do a direct query on the database or code change to let you back in - I'd suggest you contact Catalyst Europe as they'll need direct access to your system.
If you're able to let us know exactly what you did we can look at adding some code to make it impossible to lock yourself out in this way.
